In 1995, leptin was first discovered in the fat cells of a fat mouse. Its function is to tell the brain a sense of fullness, thus suppressing appetite and achieving weight loss. Matt Ozcan of Boston Children's Hospital and Harvard Medical School said that most obese people will reject leptin, and in the process, the brain no longer responds to signals from hormones that stop the diet.
The Ozcan team found that the brain cells of obese mice have increased pressure on the endoplasmic reticulum (the endoplasmic reticulum is a protein-polymerized cell structure). Because of obesity, the structure of the endoplasmic reticulum was subverted, stopped working, and began to block incoming chemical signals. This proves that stress will prevent the leptin protein response.
Subsequently, researchers tried to make the brain sensitive to leptin by reducing the pressure on the endoplasmic reticulum. They used two drugs that reduce the pressure of the endoplasmic reticulum -- tetraphenylbutyrate and sodium bovine deoxycholate. They fed any one of the drugs to obese mice fed a high-fat diet and then injected the rats with leptin. As a result, the weight of these mice dropped significantly.
The researchers said that the reduction of endoplasmic reticulum stress makes mice more sensitive to leptin, which provides new hope for the treatment of obesity. The next step will be to conduct trials on the human body.
